Selection by file type
For example, if I wanted to pull all the mp3 files out of my Stepmania songs folder to put on an mp3 disc without having space being taken up by images and dwi files, I could choose and option "select by file type" inside a folder, type in the extension, copy them, and paste them somewhere else. That way, you don't have to pull all the files out individually.
Peeling windows
I saw this on and episode of Attack of the Show!, and they mentioned it might be a nice feature to add to Longhorn (now Vista, of course). Say you have 5 Windows Explorer windows open, all covering each other. You have a file in the top window you want to move to the third one. All you do is drag the file outside that top window, then move it back towards it... the window will "peel back, exposing the second window. Repeat with the second window, and you can drop it in the third window. Awesome.
Themes
Somewhat like Firefox themes, you can skin your OS. Start button, icons, etc. would all be altered exactly to your liking.
Built-In Antivirus
The Windows Firewall works just fine, so why not have a Windows Antivirus? It'd be at least a standard to have, and you could turn it off to use a different one of your choosing (just like how someone can turn off Windows Firewall and use Norton Personal Firewall instead).
Now, maybe Windows Vista could have an extensions system (kinda like Firefox) built in to implement features like this, but that might be tricky because you'd be altering the OS's code, which could ultimately ruin your computer, or make the OS somewhat buggy. So, I'm just thinking of what could be included in the Vista's code that would be helpful in an OS that we don't have now.
Anything else you'd like to see? And please, don't just come in here and bash Windows... this is a constructive discussion. Be civilized.
