Playing on one-handed, right-hand dominant, and with the numpad on right side of keyboard (keys being 4-5-6-8), the 4321 rolls are easier to hit - than the 1234's

Right-handed. Usually depends on roll speed but as a general practice, in order of easiest to hardest:
4312/1243 (inward spins)
1234 (L to R)
4321 (R to L)
*1423/2341 (windmills/split trills)
*1324/2413 (split trills)
3421/2134 (outward spins)

*idk if they count as rolls but whatever lol

4321 is difficult. I destroy 1243 and 2134 rolls because that's what I tend to spam before a song starts.

The only real challenge would be the 1324 BS which aren't even really rolls but people like to use that pattern in what should be rolls so :shrug:

4321 are by far easier, but its more that 3->4 feels a bit hard for me. 4321, 4312, 1243 are all pretty much the same difficulty, but 1234, 2134, 3421, ect are all a bit harder. but thats assuming we aren't just playing it like a [12][34] jump trill, then all is equal.

i play different than probably everyone on here.

left hand - just index
right hand - index, middle, and ring

so going from middle to ring is a lot harder than ring to middle for me lol
i'm right handed btw

In times I've attempted spread, 1234 is easiest by far (l-->r / rh)

This is pretty accurate for my skillset as well. The "in-out" gallops or 21 and 34 are more difficult for me in general but because my right hand is much stronger I can do 1234 with relative ease at most any speeds. Now take 4321 where the left hand is doing the "harder work" and it becomes a shit-show.

12 43 or 43 12 are easiest for me, 21 34 or 34 21 are hardest

1234 is easier than 4321 which I put in the middle in terms of difficulty so my tier list is 1243 / 4312 > 1234 > 4321 > 21 34 / 34 21

ok so there seems to be a few people talking about how direction on each hand is what matters more for them (21/34 vs 12/43)

i use diagonal setup (wdko) and in-out patterns (21/34) are ever so slightly easier than out-in for me

my theory is that 12 43 type patterns are easier for people with regular setups cause its feels more natural as you wrist jack to like, flam with your longer finger (middle) first then shorter finger (index) second

with ur diagonal setup, for me just putting my fingers on like wd lp or something, it feels like my index finger becomes the longer finger rather than middle, cause of the angling, which is probably why 21 34 style rolls feel easier
